design for hMADS cells

The experimental design for human adipocyte differentiation (HAD) of hMADS cells monitors expression profiling covering confluent cells in quiescent stage, early differentiation into preadipocytes after hormonal induction, and terminal differentiation into mature adipocytes. Cells were harvested at the preconfluent stage as reference, before hormonal induction at 2 time points (-2d, 0d) and after hormonal induction at 6 time points (+8h, +1d, +2d, +5d, +10d, +15d) [Figure 1]. Expression profiling was carried out using DNA microarray technology. Three independent time series differentiation experiments were performed as biological replicates for the adipocyte differentiation of hMADS cells. Technical replicates were performed by repeating each hybridization with swapped dyes resulting in 48 microarray hybridizations for the analysis of hMADS cells adipocyte differentiation [Figure 2].
